    Methods of rapid bruise assessment and the formulation of robust bruise indices for potatoes

    When potato tubers are subjected to impacts, the sub-surface tissue may become discoloured as damaged cells produce the blue-black pigment melanin. Bruising caused during harvesting and handling can lead to downgrading of potatoes for the processing industry and quality retail trade. The two aims of this thesis were to reduce the time to detect bruising, and to develop a non-subjective method for the quantification of bruising.Reflectance spectrophotometry was investigated as a rapid, non-subjective and non- invasive way of detecting bruising. Wavelengths from ultraviolet to near infrared were selected by discriminant analysis to separate unbruised and bruised tubers. Neural nets were trained with these wavelengths to identify bruised tubers in a sample of unbruised and bruised tubers. The detection of bruising gave inconsistent results in unpeeled tubers, but proved to be reliable in peeled tubers.The rate of bruise development at air pressures up to 10 bar was measured by reflectance spectrophotometry and by a visual rating. The production of dopachrome, an orange pre-cursor pigment of melanin, was used as an early indication of bruising. Dopachrome is visible to the human eye and the time for bruise detection can be reduced to approximately 3 hours when compressed air is used.Infrared and microwave thermography were used to measure possible rises in bruised tissue temperature. Thermography was used in conjunction with scanning laser Doppler imaging to detect changes in the biological zero of bruised tissue. No significant differences could be detected between unbruised and bruised tubers using these techniques.Reflectance spectrophotometry was also used in combination with a colour digital camera to automatically measure bruise area in peeled tubers. While the camera alone could measure bruise area with precision, constant adjustments were needed. Reflectance spectrophotometry was faster but less precise than the camera for measuring bruise area.A new bruise index was formulated by taking measurements of the volume of a tuber removed per peeler stroke for the variety Record, and using the data to model the percentage volume lost for any variety. By counting the number of peeler strokes to remove a bruise an estimate of the tuber volume lost could be derived. This method can accommodate any type and size of bruise and results in an index that can be easily interpreted

    Perspectives On The Sources And Eventual Outcome Of The 2008 Economic And Financial Crisis: A Panel Discussion

    In October 2008 the Southern Utah University School of Business held a panel discussion on the current economic crisis. This discussion was part of the School’s Business Convocation series and was open to the public. The panel was designed with two components in mind. First, a pair of academics with expertise in financial institutions and business cycles offered historical and theoretical perspectives on the crisis. Second, a pair of professionals – a local banking official and a fund manager – offered perspectives on the current financial situation and practical experience based on the policy responses to past crises. As moderator, Joe Baker asked each panelist to make a short presentation on a question of general interest that was related to their area of expertise; this was followed by an open question and answer session. The participating panelists and opening questions follow. 1. Stephen Evans, Professor of Finance: Dr. Evans teaches courses on financial institutions and was asked to provide background of how the crisis occurred and what the proposed government bailout plan is expected to accomplish. 2. David Tufte, Associate Professor of Economics: Dr. Tufte is a macroeconomist and was asked to discuss the macroeconomic implications of the crisis in such areas as inflation, interest rates, economic growth and unemployment. 3. Mr. Robb Kerry, Chief Credit Officer of ADB Bank: Mr. Kerry has an extensive background in banking as a bank regulator and banker. Mr. Kerry was asked to discuss the implications of the crisis on banking credit and lending. Mr. Steve Harrop, Finance Professional in Residence: Mr. Harrop was a mutual fund manager for several decades before joining the School of Business faculty where he teaches investments and manages (pro bono) an investment fund. Mr. Harrop will discuss the implications of the crisis on the stock and bond markets

    Oxidation of tertiary amine-derivatized surfaces to control protein adhesion

    Selective oxidation of omega-tertiary amine self-assembled thiol monolayers to tertiary amine N-oxides is shown to transform the adhesion of model proteins lysozyme and fibrinogen upon them. Efficient preparation of both secondary and tertiary linker amides as judged by X-ray photoelectron spectroscopy (XPS) and water droplet contact angle was achieved with an improved amide bond formation on gold quartz crystal microbalance (QCM) sensors using 2-(1H-7-azabenzotriazol-1-yl)-1,1,3,3-tetramethyl hexafluorophosphate methanaminium uronium (HATU). Oxidation with hydrogen peroxide was similarly assessed, and adhesion of lysozyme and fibrinogen from phosphate buffered saline was then assayed by QCM and imaged by AFM. Tertiary amine-functionalized sensors adsorbed multilayers of aggregated lysozyme, whereas tertiary amine N-oxides and triethylene glycol-terminated monolayers are consistent with small protein aggregates. The surface containing a dimethylamine N-oxide headgroup and ethyl secondary amide linker showed the largest difference in adsorption of both proteins. Oxidation of tertiary amine decorated surfaces therefore holds the potential for selective deposition of proteins and cells through masking and other patterning techniques

    Recent results on KS0^0_{\rm S} and hyperon production in Pb-Pb collisions at 40 and 158 AA GeV/cc beam momentum from the NA57 experiment at CERN SPS are presented. Yields and ratios are compared with those measured by the NA49 experiment, where available.     Synthesis and characterisation of halide, separated ion pair, and hydride cyclopentadienyl iron bis(diphenylphosphino)ethane derivatives

    Treatment of anhydrous FeX₂ (X = Cl, Br, I) with one equivalent of bis(diphenylphosphino)ethane (dppe) in refluxing THF afforded analytically pure white (X = Cl), light green (X = Br), and yellow (X = I) [FeX₂(dppe)]n (X = Cl, I; Br, II; I, III). Complexes I–III are excellent synthons from which to prepare a range of cyclopentadienyl derivatives. Specifically, treatment of I–III with alkali metal salts of C₅H₅ (Cp, series 1), C₅Me₅ (Cp*, series 2), C₅H₄SiMe₃ (Cp′, series 3), C₅H₃(SiMe₃)₂ (Cp′′, series 4), and C₅H₃(But)₂ (Cptt, series 5) afforded [Fe(Cp†)(Cl)(dppe)] 1Cl–5Cl, [Fe(Cp†)(Br)(dppe)] 1Br–5Br, and [Fe(Cp†)(I)(dppe)] 1I–5I (Cp† = Cp, Cp*, Cp′, Cp′′, or Cptt). Dissolution of 1I–5I in acetonitrile, or treatment of 1Cl–5Cl with Me₃SiI in acetonitrile (no halide exchange reactions were observed in other solvents) afforded the separated ion pair complexes [Fe(Cp†)(NCMe)(dppe)][I] 1SIP–5SIP. Attempts to reduce 1Cl–5Cl, 1Br–5Br, and 1I–5I with a variety of reductants (Li-Cs, KC₈, Na/Hg) were unsuccessful. Treatment of 1Cl–5Cl with LiAlH₄ gave the hydride derivatives [Fe(Cp†)(H)(dppe)] 1H–5H. This report provides a systematic account of reliable methods of preparing these complexes which may find utility in molecular wire and metal–metal bond chemistries. The complexes reported herein have been characterised by X-ray diffraction, NMR, IR, UV/Vis, and Mössbauer spectroscopies, cyclic voltammetry, density functional theory calculations, and elemental analyses, which have enabled us to elucidate the electronic structure of the complexes and probe the variation of iron redox properties as a function of varying the cyclopentadienyl or halide ligand

    Visualization of replication initiation and elongation in Drosophila

    Chorion gene amplification in the ovaries of Drosophila melanogaster is a powerful system for the study of metazoan DNA replication in vivo. Using a combination of high-resolution confocal and deconvolution microscopy and quantitative realtime PCR, we found that initiation and elongation occur during separate developmental stages, thus permitting analysis of these two phases of replication in vivo. Bromodeoxyuridine, origin recognition complex, and the elongation factors minichromosome maintenance proteins (MCM)2–7 and proliferating cell nuclear antigen were precisely localized, and the DNA copy number along the third chromosome chorion amplicon was quantified during multiple developmental stages. These studies revealed that initiation takes place during stages 10B and 11 of egg chamber development, whereas only elongation of existing replication forks occurs during egg chamber stages 12 and 13. The ability to distinguish initiation from elongation makes this an outstanding model to decipher the roles of various replication factors during metazoan DNA replication. We utilized this system to demonstrate that the pre–replication complex component, double-parked protein/cell division cycle 10–dependent transcript 1, is not only necessary for proper MCM2–7 localization, but, unexpectedly, is present during elongation

    This paper reviews and examines emerging academic approaches to the study of ‘sexualized culture’; an examination made necessary by contemporary preoccupations with sexual values, practices and identities, the emergence of new forms of sexual experience and the apparent breakdown of rules, categories and regulations designed to keep the obscene at bay. The paper maps out some key themes and preoccupations in recent academic writing on sex and sexuality, especially those relating to the contemporary or emerging characteristics of sexual discourse. The key issues of pornographication and democratization, taste formations, postmodern sex and intimacy, and sexual citizenship are explored in detail. </p
